The influence of dilution solution and storage time on the quality of semen in Tam Hoang chickens

Abstract

The study was conducted to evaluate the effects of extenders and storage time on the semen quality of Tam Hoang roosters. Semen from 20 Tam Hoang roosters aged 6-8 months, raised at the experimental farm of Tay Do University, was used. The research took place from August 2023 to March 2024, with semen collection every three days using the abdominal massage method. The semen was stored in two types of extenders, A and B, at 0, 4, 6, and 12 hours in a refrigerator at 4°C. Sperm characteristics and quality parameters were recorded by visual observation, electron microscopy, and video recording. The analysis showed that the semen was 100% milky white. When stored in extender A at 0 hours, sperm had fast motility (75%) and progressive motility (70%), while in extender B, sperm had fast motility (18%), medium motility (50%), and progressive motility (60%). After 12 hours of storage, sperm in extender A had medium and slow motility rates of 55% and 31%, compared to 45% and 50% in extender B. There was a statistically significant difference between the two extenders in terms of pH, motility, abnormality rate, and sperm viability (P<0.05). Extender B had a higher pH, while sperm motility, abnormality rate, and viability were lower compared to extender A. Similarly, sperm motility, viability, and concentration decreased significantly, while the abnormality rate progressively increased over the storage times of 0, 4, 6, and 12 hours. Extender A was superior to extender B in terms of semen quality. Additionally, the 0-hour storage time showed the best sperm quality, which gradually decreased over time. A strong positive correlation (0.96) was found between semen volume and sperm concentration, while a negative correlation was observed between abnormality rate and sperm viability (-0.455), and between concentration and abnormality rate (-0.387).
